The molecule you're making right now
Most supplements in the longevity aisle are strangers — molecules you'd never meet without a bottle. Alpha-ketoglutarate is the opposite. Right now, in every cell in your body, it's being made and used up thousands of times a second. It's a linchpin of the Krebs cycle (also called the citric-acid or TCA cycle), the elegant loop of reactions that pulls energy out of the food you eat. Chemically it's nothing glamorous: a small five-carbon acid, C5H6O5, that biochemistry students draw on exam papers. You are, quite literally, powered by the stuff.
So why is a molecule from your own metabolism suddenly one of 2026's most-searched supplements? Because AKG turns out to moonlight. Beyond shuttling energy, it acts as a signalling molecule and a required co-factor for a family of enzymes that edit your epigenome and tune your immune system.[4] And here's the hook that launched a thousand headlines: the amount of AKG circulating in your blood drops steeply with age — by some estimates as much as tenfold between youth and old age. Naturally, someone asked the obvious question: what happens if you top it back up? That's exactly the kind of “refill the tank of youth” claim a clean-longevity site is obligated to walk over and poke with a stick.
The mouse data that started it
The excitement is not built on nothing. In 2020, a team at the Buck Institute for Research on Aging published a genuinely eye-catching result in Cell Metabolism: feeding aging mice calcium AKG (CaAKG) didn't just make them live modestly longer — it made them live better, right up to the end.[1] The treated mice were measurably less frail: shinier coats, better gait, less of the physical decline that normally precedes death. In the jargon, AKG produced a “compression of morbidity” — more of life spent healthy, less of it spent falling apart. If you had to pick the single most desirable outcome in all of longevity science, that would be a strong contender.
And the researchers went further than “it worked” — they found a plausible why. AKG appeared to ramp up an anti-inflammatory signal called IL-10, which in turn quieted the systemic, smouldering inflammation that quietly ages tissues (the phenomenon gerontologists have nicknamed “inflammaging”). That mechanism rhymes nicely with a lot of other longevity biology, from fasting to the senolytic drugs that clear inflammatory “zombie” cells. A separate group added another thread the same year, showing in Nature Communications that AKG rebuilt bone mass in aged mice by reversing age-related changes to how their DNA was packaged.[2] Two independent labs, two different tissues, one consistent story. Lovely for the mice. But you, dear reader, are not a mouse, so let's see what survives the jump to humans.
The 8-years-younger headline — and the fine print
Here's the study behind every AKG headline you've seen. In 2021, researchers reported in the journal Aging that 42 people who had taken a commercial AKG-based supplement called Rejuvant for an average of seven months showed an average drop of almost eight years in their biological age, measured with a TruAge DNA-methylation clock.[3] Eight years. From a pill. You can see why that number went around the internet at the speed of light.
Now the part the headlines quietly leave out — and on this site there's always a part they leave out. First, the study was retrospective: the researchers looked back at people who had already chosen to take the supplement, rather than randomly assigning it. That's a magnet for bias, because the kind of person who buys a longevity supplement and diligently gets their DNA methylation tested is rarely also a chain-smoking couch potato. Second, there was no placebo group at all — nothing to compare against, so we can't know how much of that eight years is AKG and how much is regression to the mean, lifestyle, or the noise in the clock itself. Third, the sample was tiny (42 people) and self-selected. Fourth, Rejuvant isn't pure AKG — it's calcium AKG blended with vitamins A and D, so even if the effect is real, you can't cleanly attribute it to AKG. And fifth, in the interest of full disclosure: the study was conducted and authored by people connected to the companies behind the product and the test. None of that makes the result wrong. It makes it a hypothesis wearing a very persuasive number.
What the honest human evidence actually says
Strip away the one splashy study and what's left is thinner and older than the marketing implies. AKG has a genuinely long history in human nutrition — but for muscle recovery, wound healing and helping patients bounce back after surgery, studies mostly from the 1980s and 1990s.[4] A 2021 review from Brian Kennedy's longevity group in Singapore laid it out plainly: AKG's effects on aging are well demonstrated in worms and mice, biologically plausible in people, and — as of that writing — essentially untested in a modern, rigorous human aging trial. That's the honest state of play. A great mechanism and a great mouse are not a great randomized human result, and it's precisely the gap where a dozen other longevity darlings have quietly stumbled, from the taurine hype cycle to NMN's yawning gap between raising a biomarker and actually slowing aging.
It's also worth being clear-eyed about biological-age clocks themselves. A DNA-methylation age is a statistical estimate, not a readout on a dashboard, and the numbers can wobble from one blood draw to the next for reasons that have nothing to do with your supplement cabinet. When a headline says a pill made someone “eight years younger,” what it really means is the clock's estimate moved — which is interesting, and not the same as adding eight years to their life. So why is it a 2026 story?
Fair question — if the human proof is thin, why is AKG trending now? Two reasons, one good and one to watch. The good one: the science is finally being done properly. Kennedy's Singapore group is running a trial called ABLE — a genuine double-blind, placebo-controlled randomized study giving 120 middle-aged adults either 1 gram a day of sustained-release calcium AKG or a placebo, specifically recruiting people whose biological age is already running ahead of their birthday.[5] Its main question is exactly the one the viral study couldn't answer: does AKG actually lower DNA-methylation age versus a dummy pill? That is the readout worth waiting for, and it's why AKG keeps resurfacing in longevity feeds this year.
The reason to keep your guard up is the mirror image: “refill the molecule your body loses with age and turn back your clock” is an almost perfect marketing story, and the supplement industry knows it. A falling blood level with age plus one dramatic before-and-after number is catnip for ad copy. The molecule is real, the mouse data are real, the human proof is not yet real — and that ordering matters enormously.
The clean verdict
Alpha-ketoglutarate is one of the more genuinely plausible longevity compounds we cover, and we don't hand that out lightly. It has a real, central role in your metabolism, striking lifespan-and-healthspan data in mice, a coherent anti-inflammatory mechanism, and — unusually — a proper randomized human trial already running to test it. That's a stronger foundation than most of the anti-aging aisle can claim. What it does not yet have is the one thing that would justify the “reverses aging” headlines: a placebo-controlled human trial showing the effect is real. The famous eight-years-younger number rests on a study too weak to carry it.
So here's the clean framing. If you're intrigued and want to run a personal experiment with roughly 1 gram a day of calcium AKG, it's biologically defensible and appears well tolerated — just go in knowing you're funding an experiment, not buying a proven result, and don't let a shiny capsule distract you from the boring stuff that actually moves biological age. Because the unglamorous truth we keep circling back to is that no metabolite in a bottle beats sleep, muscle, cardio and a decent plate of food. Alpha-ketoglutarate (formula C5H6O5) is a natural metabolite your cells already make as a central intermediate of the Krebs (TCA) cycle — the reaction chain that turns food into cellular energy.[4] Beyond energy, it acts as a signalling molecule and a co-factor for enzymes that regulate the epigenome and inflammation. Supplements usually deliver it as calcium AKG (Ca-AKG), because the free acid is unstable.
In mice, calcium AKG extended lifespan and reduced frailty, largely by dialling down chronic inflammation.[1] In humans the headline comes from a 2021 retrospective study of 42 people taking the AKG-plus-vitamins formula Rejuvant, who showed an average 8-year drop in DNA-methylation age.[3] But that study had no placebo group, a small self-selected sample, and was run by the product's makers — so it's a promising signal, not proof. The placebo-controlled ABLE trial is what will actually settle it.[5]
AKG is a molecule your body makes constantly, and older human studies using it for muscle recovery, wound healing and post-surgery nutrition reported good tolerability.[4] Modern longevity trials use around 1 gram per day of sustained-release calcium AKG.[5] Long-term safety data in healthy people are still limited, so check with a doctor first — especially if you're pregnant, on medication, or managing a health condition.
